This variant lies in the GLA gene (transcript NM_000169.3) at coding-DNA position 36, where C is replaced by A; at the protein level this means converts the codon for cysteine at residue 12 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: GLA p.Cys12Ter (c.36C>A) is a nonsense variant that introduces a premature stop codon at amino acid position 12, creating a truncated protein that may be subject to nonsense-mediated mRNA decay. This variant has been observed in at least one proband affected with Fabry disease (PMID:28087245;36140787). The variant was found to segregate with disease in at least one affected family (PMID:28087245). Functional studies have been reported; however, the significance of the findings remain unclear and/or they were performed in patient cells (PMID:36140787;28087245;30871880). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify GLA p.Cys12Ter (c.36C>A) as a pathogenic variant.
