Submitter rationale: GLA p.Met187_Ser188dup (c.559_564dup) is an in-frame duplication variant that results in the duplication of two amino acids, Methionine at position 187 and Serine at position 188. To our knowledge, this variant has not been reported in patients affected with Fabry disease in the published literature. At least one functional study has demonstrated a substantial alteration in protein function relative to the wild-type (PMID:27657681). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify GLA p.Met187_Ser188dup (c.559_564dup) as a likely pathogenic variant.
