This variant lies in the GLA gene (transcript NM_000169.3) at coding-DNA position 1098, deleting one base; at the protein level this means shifts the reading frame starting at isoleucine residue 367, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: GLA p.Ile367SerfsTer24 (c.1098del) is a frameshift variant that results in the production of a truncated protein. This variant has been observed in at least one proband affected with Fabry disease (PMID:27560961). The variant was found to segregate with disease in at least one affected family (PMID:27560961). Functional studies have been reported; however, the significance of the findings remain unclear and/or were performed in patient cells (PMID:27560961). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify GLA p.Ile367SerfsTer24 (c.1098del) as a pathogenic variant.
