Submitter rationale: The p.R1671C variant (also known as c.5011C>T), located in coding exon 35 of the SMARCA4 gene, results from a C to T substitution at nucleotide position 5011. The arginine at codon 1671 is replaced by cysteine, an amino acid with highly dissimilar properties. This variant has been detected in multiple individuals with no reported features of Coffin-Sirris syndrome (Ambry internal data). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive.
