Submitter rationale: The p.S141L variant (also known as c.422C>T), located in coding exon 3 of the SMARCA4 gene, results from a C to T substitution at nucleotide position 422. The serine at codon 141 is replaced by leucine, an amino acid with dissimilar properties. This variant has been detected in multiple individuals with no reported features of Coffin-Siris syndrome (Ambry internal data).
