Likely pathogenic for Euthyroid goiter — the classification assigned by Center of Human Genetics, Hôpital Erasme to NM_177438.3(DICER1):c.2988-3C>G, citing ACMG Guidelines, 2015: The c.2988-3C>G variant is absent from the gnomAD v4 control database and is not described in the literature. The three prediction software programs tested support an impact on splicing (SpliceAI: Splice-Altering / strong (0.91), dbscSNV Ada: Deleterious (1), and dbscSNV RF: Deleterious (0.77)). Family segregation was performed on 4 patients (three generations) of the same family. All of them carry this DICER1 variant and suffer from multinodular goiter and two of them have had nephrectomy in childhood. For these reasons, the variant was classified as a likely pathogenic variant (class IV).
