Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001845.6(COL4A1):c.955C>A (p.Gln319Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine, which is neutral and polar, with lysine, which is basic and polar, at codon 319 of the COL4A1 protein (p.Gln319Lys). This variant is present in population databases (no rsID available, gnomAD 0.0009%). This variant has not been reported in the literature in individuals affected with COL4A1-related conditions. An algorithm developed to predict the effect of missense changes on protein structure and function outputs the following: PolyPhen-2: "Not Available". The lysine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
