Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_005732.4(RAD50):c.3244G>A (p.Glu1082Lys), citing Ambry Variant Classification Scheme 2023: The p.E1082K variant (also known as c.3244G>A), located in coding exon 21 of the RAD50 gene, results from a G to A substitution at nucleotide position 3244. The glutamic acid at codon 1082 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
