This variant lies in the PKD1 gene (transcript NM_001009944.3) at coding-DNA position 9590, where T is replaced by G; at the protein level this means replaces leucine at residue 3197 with arginine — a missense variant. Submitter rationale: The variant is not observed in the gnomAD v4.1.0 dataset. Predicted Consequence/Location: Missense variant In silico tool predictions suggest damaging effect of the variant on gene or gene product [REVEL: 0.74 (>=0.6, sensitivity 0.68 and specificity 0.92)]. The same nucleotide change resulting in the same amino acid change has been previously reported to be associated with PKD1-related disorder (PMID: 37509056). A different missense change at the same codon (p.Leu3197Pro) has been reported to be associated with PKD1-related disorder (PMID: 33437033). However the evidence of pathogenicity is insufficient at this time. Therefore, this variant is classified as VUS according to the recommendation of ACMG/AMP guideline.
