NM_025077.4(TOE1):c.373_374del (p.Leu125fs) was classified as Likely pathogenic by Genetic Services Laboratory, University of Chicago, citing ACMG Guidelines, 2015: DNA sequence analysis of the TOE1 gene demonstrated a 2 base pair deletion in exon 5, c.373_374del. This sequence change results in an amino acid frameshift and creates a premature stop codon 35 amino acids downstream of the change, p.Leu125Alafs*35. This sequence change is predicted to result in an abnormal transcript, which may be degraded, or may lead to the production of a truncated TOE1 protein with potentially abnormal function. While this particular deletion has not previously been described in the literature, other deletions in the TOE1 gene have been described in several individuals with TOE1-related disorders (PMIDs: 28092684). This sequence change has been described in one individual the gnomAD database, corresponding to an overall population frequency of 0.0004% (dbSNP rs770878310). Taken together, the available evidence indicates that this sequence change is likely pathogenic.
