NM_001252.5(CD70):c.371A>G (p.His124Arg) was classified as Uncertain significance by Genetic Services Laboratory, University of Chicago, citing ACMG Guidelines, 2015: DNA sequence analysis of the CD70 gene demonstrated a sequence change, c.371A>G, in exon 3 that results in an amino acid change, p.His124Arg. This sequence change does not appear to have been previously described in individuals with CD70-related disorders and has also not been described in population databases such as ExAC and gnomAD. The p.His124Arg change affects a poorly conserved amino acid residue located in a domain of the CD70 protein that is known to be functional. The p.His124Arg substitution appears to be benign using several in-silico pathogenicity prediction tools (SIFT, PolyPhen2, Align GVGD, REVEL). Due to insufficient evidences and the lack of functional studies, the clinical significance of the p.His124Arg change remains unknown at this time.
