This variant lies in the FANCA gene (transcript NM_000135.4) at coding-DNA position 661, where A is replaced by G; at the protein level this means replaces methionine at residue 221 with valine — a missense variant. Submitter rationale: Variant summary: FANCA c.661A>G (p.Met221Val) results in a conservative amino acid change located in the Fanconi anaemia group A protein, N-terminal domain (IPR031729) of the encoded protein sequence. Five of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 3.2e-05 in 249370 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.661A>G has been reported in the literature in an individual affected with Pancreatic cancer (Rogers_2004). These report(s) do not provide unequivocal conclusions about association of the variant with Fanconi Anemia. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported.
