Uncertain significance for Malignant hyperthermia, susceptibility to, 1 — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_000540.3(RYR1):c.296C>T (p.Thr99Met), citing ACMG Guidelines, 2015: This missense variant replaces threonine with methionine at codon 99 of the RYR1 protein. Variant occurs in the N-terminal region (a.a. 1-552) of RYR1 that is considered to be a hotspot for pathogenic variants that contribute to malignant hyperthermia susceptibility (PMID: 21118704). Computational prediction suggests that this variant may have deleterious impact on protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has not been reported in individuals affected with RYR1-related disorders in the literature. This variant has been identified in 10/251164 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
