Pathogenic for Fabry disease — the classification assigned by Genomenon, Inc, Genomenon, Inc to NM_000169.3(GLA):c.736del (p.Thr246fs), citing Genomenon Sequence Variant Interpretation Standards. This variant lies in the GLA gene (transcript NM_000169.3) at coding-DNA position 736, deleting one base; at the protein level this means shifts the reading frame starting at threonine residue 246, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: GLA p.Thr246HisfsTer23 (c.736del) is a frameshift variant that results in the production of a truncated protein which is predicted to undergo nonsense-mediated mRNA decay. This variant has been observed in at least one proband affected with Fabry disease (PMID:35743707). The variant was found to segregate with disease in at least one affected family (PMID:35743707). It is absent or not present at a significant frequency in gnomAD. In conclusion, we classify GLA p.Thr246HisfsTer23 (c.736del) as a pathogenic variant.
