Pathogenic for Familial cancer of breast; Fanconi anemia complementation group J — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_032043.3(BRIP1):c.2786_2789dup (p.Pro931fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. While this particular variant has not been reported in the literature, loss-of-function variants in BRIP1 are known to be pathogenic (PMID: 16116423, 17033622, 21964575). This sequence change inserts 4 nucleotides in exon 19 of the BRIP1 mRNA (c.2786_2789dupTATC), causing a frameshift at codon 931. This creates a premature translational stop signal (p.Pro931Ilefs*9) and is expected to result in an absent or disrupted protein product.
