This variant is absent from gnomAD v2.1.1 and v4.1.0 (PM2_Supporting). This variant is located within the HNF6/OC2 binding site of the promoter (c.-132 to c.-141) of HNF4A, which is defined as critical for the protein’s function by the ClinGen MDEP (PM1_Supporting). This variant was identified in an individual with diabetes; however, the calculated MODY probability is <50%, therefore PP4 will not be applied (internal lab contributors). In summary, c.-136A>C meets the criteria to be classified as a variant of uncertain significance for monogenic diabetes.
