(Genet Med. 2024): The classification of the A allele of rs965513 as an 'established risk allele' for papillary thyroid carcinoma is based on consistent evidence from multiple sources: (a) Case-Control Data: In our cohort, the frequency of the A allele was significantly higher in the papillary thyroid carcinoma patient group (13.3%) compared to the control group (5.9%). (b) Replicated Association: This association has been consistently replicated in multiple large-scale genome-wide association studies (GWAS) across diverse populations, establishing it as a well-known risk locus for the disease (e.g., Gudmundsson 2009, Jones 2012, Tcheandjieu 2016, Owens 2021). (c) Database Records: This variant is curated as a risk allele in authoritative public databases such as the GWAS Catalog and dbSNP. The strength, consistency, and reproducibility of the association evidence across multiple studies and in our own data meet the criteria for an 'established risk allele' as defined by the ClinGen Risk Allele Working Group recommendations (PMID: 38054408).
