Uncertain significance for SKIN/HAIR/EYE PIGMENTATION 5, BLACK/NONBLACK HAIR; Oculocutaneous albinism type 4 — the classification assigned by Laboratory of Genetic Epidemiology, Research Centre for Medical Genetics to NC_000005.10:g.33984646_33984746del, citing ACMG Guidelines, 2015: The deletion of 101 bp, affecting the 5'UTR and promoter region of the SLC45A2 gene, was identified in a compound heterozygous state in a proband diagnosed with albinism in trans with previously published pathogenic variant NM_016180.4:c.264delC, which leading to f the formation of a premature stop codon p.(Gly89AspfsTer24). This deletion truncates the 5′-UTR and promoter region of SLC45A2, potentially disrupting its transcription. It is not listed in gnomAD v2.1.1. Taken together, the variant meets the following ACMG/AMP criteria and can be classified as uncertain significance with PM2, PM3, PP4 criteria.
