Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.2263T>C (p.Cys755Arg), citing Ambry Variant Classification Scheme 2023: The p.C755R variant (also known as c.2263T>C), located in coding exon 14 of the ATM gene, results from a T to C substitution at nucleotide position 2263. The cysteine at codon 755 is replaced by arginine, an amino acid with highly dissimilar properties. In an assay testing ATM function, this variant showed a functionally normal result (Lee KS et al. Cell, 2025 Sep;188:5081-5099.e27). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
