NM_000051.4(ATM):c.8240A>G (p.Lys2747Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K2747R variant (also known as c.8240A>G), located in coding exon 55 of the ATM gene, results from an A to G substitution at nucleotide position 8240. The lysine at codon 2747 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
