Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.6959A>G (p.Asp2320Gly), citing Ambry Variant Classification Scheme 2023. This variant lies in the ATM gene (transcript NM_000051.4) at coding-DNA position 6959, where A is replaced by G; at the protein level this means replaces aspartic acid at residue 2320 with glycine — a missense variant. Submitter rationale: The p.D2320G variant (also known as c.6959A>G), located in coding exon 46 of the ATM gene, results from an A to G substitution at nucleotide position 6959. The aspartic acid at codon 2320 is replaced by glycine, an amino acid with similar properties. In an assay testing ATM function, this variant showed a functionally normal result (Lee KS et al. Cell, 2025 Sep;188:5081-5099.e27). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
