Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.9006C>A (p.Phe3002Leu), citing Ambry Variant Classification Scheme 2023: The p.F3002L variant (also known as c.9006C>A), located in coding exon 62 of the ATM gene, results from a C to A substitution at nucleotide position 9006. The phenylalanine at codon 3002 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
