Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.2915C>T (p.Pro972Leu), citing Ambry Variant Classification Scheme 2023: The p.P972L variant (also known as c.2915C>T), located in coding exon 18 of the ATM gene, results from a C to T substitution at nucleotide position 2915. The proline at codon 972 is replaced by leucine, an amino acid with similar properties. This variant was reported in 1 of 701 Brazilian individuals with features consistent with a hereditary breast and/or ovarian cancer syndrome (Faria JP et al. Breast Cancer Res Treat, 2024 Oct;207:615-624). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
