NM_000051.4(ATM):c.8561G>A (p.Arg2854His) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): This variant is denoted ATM c.8561G>A at the cDNA level, p.Arg2854His (R2854H) at the protein level, and results in the change of an Arginine to a Histidine (CGC>CAC). This variant has not, to our knowledge, been published in the literature as pathogenic or benign. ATM Arg2854His was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. Since Arginine and Histidine share similar properties, this is considered a conservative amino acid substitution. ATM Arg2854His occurs at a position that is conserved across species and is located within the PI3K/PI4K kinase domain (Tavtigian 2009, Stracker 2013). In silico analyses predict that this variant is probably damaging to protein structure and function. Based on currently available information, it is unclear whether ATM Arg2854His is pathogenic or benign. We consider it to be a variant of uncertain significance.
