Likely pathogenic — the classification assigned by GeneDx to NM_002755.4(MAP2K1):c.730T>G (p.Ser244Ala), citing GeneDx Variant Classification (06012015). This variant lies in the MAP2K1 gene (transcript NM_002755.4) at coding-DNA position 730, where T is replaced by G; at the protein level this means replaces serine at residue 244 with alanine — a missense variant. Submitter rationale: The S244A variant has not been published as a pathogenic variant, nor has it been reported as a benign variant to our knowledge. The S244A variant is not observed in large population cohorts (Lek et al., 2016; 1000 Genomes Consortium et al., 2015; Exome Variant Server). The S244A variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. This substitution occurs at a position that is conserved across species. In silico analysis predicts this variant is probably damaging to the protein structure/function. This variant has been observed de novo in a patient tested at GeneDx.
