NM_006206.6(PDGFRA):c.1624C>T (p.Leu542Phe) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L542F variant (also known as c.1624C>T), located in coding exon 10 of the PDGFRA gene, results from a C to T substitution at nucleotide position 1624. The leucine at codon 542 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
