NM_004260.4(RECQL4):c.3104dup (p.His1036fs) was classified as Pathogenic for familial breast cancer- squamous cell carcinoma; Familial cancer of breast by Department of Genetics and Molecular Biology, Isfahan University of Medical Sciences, citing ACMG Guidelines, 2015. This variant lies in the RECQL4 gene (transcript NM_004260.4) at coding-DNA position 3104, duplicating one base; at the protein level this means shifts the reading frame starting at histidine residue 1036, producing a truncated or aberrant protein — a frameshift variant. Submitter rationale: Analyzing the data obtained from ES identified a heterozygous frameshift variant (c.3104_3105insT, p.F1035Lfs*12) in exon 18 of the RECQL4 gene (NM_004260.4). This one nucleotide insertion causes termination of translation at amino acid 1046 rather than 1209, which losses its catalytic competent. The pathogenicity effects of this variant were confirmed according to the ACMG guidelines (met PVS1, PS1, PM1, PM2 and PM4 criteria).
