Uncertain significance for Hypertrophic cardiomyopathy — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000256.3(MYBPC3):c.1624+206_1624+207ins236, citing Invitae Variant Classification Sherloc (09022015): This sequence change is an insertion in intron 17 of 236 nucleotides of the MYBPC3 mRNA (c.1624+206_1624+207ins236), The inserted nucleotides are identical to the area surrounding the coding 3' region of exon 23, and the first 101 nucleotides of intron 23 of MYBPC3 (c.2174_2308+101). This variant is not present in population databases (ExAC no frequency) and has not been reported in the literature in individuals with a MYBPC3-related disease. Algorithms developed to predict the effect of sequence changes on mRNA splicing suggest that this variant may alter mRNA splicing, but this prediction has not been confirmed by published transcriptional studies. In summary, this is a novel, deep-intronic insertion of the 3' region of exon 23 which has an uncertain effect on splicing of the MYBPC3 mRNA. For these reasons, it has been classified as a Variant of Uncertain Significance.
