Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_002317.7(LOX):c.640G>A (p.Asp214Asn), citing Ambry Variant Classification Scheme 2023: The p.D214N variant (also known as c.640G>A), located in coding exon 2 of the LOX gene, results from a G to A substitution at nucleotide position 640. The aspartic acid at codon 214 is replaced by asparagine, an amino acid with highly similar properties. This variant was reported in individual(s) with features consistent with thoracic aortic aneurysm and dissection (Ambry internal data). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
