A frameshift deletion in LACTB (NM_001288585.2), exon 4: c.785_786delTT, resulting in a frameshift at phenylalanine 262 and a premature stop codon two residues downstream (p.F262Yfs*2). To our knowledge, functional studies specific to this variant have not been reported. This variant has not been described in individuals with Familial Hypercholesterolemia (FH) in the literature; however, the LACTB gene is associated with lipid metabolism according to the LIPID MAPS Proteome Database (LMPD). Computational predictions using MutationTaster and PolyPhen-2 suggest that this variant is likely deleterious and possibly damaging to protein function (GRCh38).
