D. Hinduja Hospital & MRC: This variant was identified as part of the ICMR-funded project (Ref No. 2020-3881). A frameshift deletion in AGPAT5 (NM_018361.5), exon 7: c.858delA, causing a lysine-to-arginine substitution at codon 290 and introducing a premature stop codon 4 residues downstream (p.K290Rfs*4). To our knowledge, functional studies specific to this variant have not been reported. This variant has not been described in individuals with Familial Hypercholesterolemia (FH) in the literature; however, the AGPAT5 gene is associated with lipid metabolism according to the LIPID MAPS Proteome Database (LMPD). Computational predictions using MutationTaster and PolyPhen-2 suggest that this variant is likely deleterious and possibly damaging to protein function (GRCh38).
