A missense variant in INPP4B (NM_001101669.3), exon 22: c.2267T>C, leading to a leucine-to-proline substitution at position 756 (p.L756P) To our knowledge, functional studies specific to this variant have not been reported. This variant has not been described in individuals with Familial Hypercholesterolemia (FH) in the literature; however, the INPP4B gene is associated with lipid metabolism according to the LIPID MAPS Proteome Database (LMPD). Computational predictions using MutationTaster and PolyPhen-2 suggest that this variant is likely deleterious and possibly damaging to protein function (GRCh38).
