Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Sema4, Sema4 to NM_004260.4(RECQL4):c.2756-6C>T, citing Sema4 Curation Guidelines: The RECQL4 c.2756-6C>T variant has not been reported in the literature to our knowledge. This variant was observed in 6/126226 chromosomes in the Non-Finnish European population according to the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). This variant has been reported in ClinVar (Variation ID: 407014). In silico tools do not suggest negative effect on normal splicing, though these predictions have not been confirmed by functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
