NM_000465.4(BARD1):c.1805G>T (p.Ser602Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S602I variant (also known as c.1805G>T), located in coding exon 8 of the BARD1 gene, results from a G to T substitution at nucleotide position 1805. The serine at codon 602 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
