Submitter rationale: The p.D105E variant (also known as c.315C>A), located in coding exon 2 of the CDKN2A gene, results from a C to A substitution at nucleotide position 315. The aspartic acid at codon 105 is replaced by glutamic acid, an amino acid with highly similar properties. Of note, this alteration is also known as c.358C>A (p.R120S)in the p14(ARF) isoform. This alteration has been reported in the germline of an individual diagnosed with a sporadic cutaneous melanoma (Fujimoto A et al. Oncogene. 1999 Apr;18:2527-32). This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
