Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004360.5(CDH1):c.2122C>A (p.Pro708Thr), citing Ambry Variant Classification Scheme 2023: The p.P708T variant (also known as c.2122C>A), located in coding exon 13 of the CDH1 gene, results from a C to A substitution at nucleotide position 2122. The proline at codon 708 is replaced by threonine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
