Uncertain significance for Hereditary spastic paraplegia 11 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_025137.4(SPG11):c.5269C>T (p.His1757Tyr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine with tyrosine at codon 1757 of the SPG11 protein (p.His1757Tyr). The histidine residue is weakly conserved and there is a moderate physicochemical difference between histidine and tyrosine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with SPG11-related disease. ClinVar contains an entry for this variant (Variation ID: 406511).
