NM_000492.4(CFTR):c.821C>G (p.Ala274Gly) was classified as Uncertain significance for Cystic fibrosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.A274G variant (also known as c.821C>G), located in coding exon 7 of the CFTR gene, results from a C to G substitution at nucleotide position 821. The alanine at codon 274 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
