NM_000335.5(SCN5A):c.2570G>A (p.Gly857Asp) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G857D variant (also known as c.2570G>A), located in coding exon 15 of the SCN5A gene, results from a G to A substitution at nucleotide position 2570. The glycine at codon 857 is replaced by aspartic acid, an amino acid with similar properties, and is located in the transmembrane DII-S5 domain. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
