Submitter rationale: ABCB11 p.Ile1061ValfsTer35 (c.3181_3184del) is a frameshift variant that results in the production of a truncated protein which is predicted to undergo nonsense-mediated mRNA decay. This variant has been observed in at least one proband with an ABCB11-related disorder (PMID:35626323;20232290). The variant was found to segregate with disease in at least one affected family (PMID:20232290). It is absent or not present at a significant frequency in gnomAD.
