NM_000548.5(TSC2):c.1946+5G>T was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Sema4, Sema4, citing Sema4 Curation Guidelines: To the best of our knowledge, the TSC2 c.1946+5G>T has not been reported in individuals with TSC2-related disease. It was not observed in the large and broad cohorts of the Genome Aggregation Database (http://gnomad.broadinstitute.org, PMID: 32461654). The variant has been reported in ClinVar (Variation ID: 406022). This nucleotide position is highly conserved in available vertebrate species. In silico tools suggest the impact of the variant on RNA splicing is deleterious, though these predictions have not been confirmed by functional studies. The evidence is insufficient to meet ACMG/AMP criteria for classifying the variant as benign or pathogenic. Thus, the clinical significance of this variant is currently uncertain.
