NM_006231.4(POLE):c.3257G>A (p.Gly1086Asp) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G1086D variant (also known as c.3257G>A), located in coding exon 26 of the POLE gene, results from a G to A substitution at nucleotide position 3257. The glycine at codon 1086 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
