NM_020975.6(RET):c.178C>A (p.Pro60Thr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.P60T variant (also known as c.178C>A), located in coding exon 2 of the RET gene, results from a C to A substitution at nucleotide position 178. The proline at codon 60 is replaced by threonine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
