Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001354604.2(MITF):c.670G>C (p.Asp224His), citing Ambry Variant Classification Scheme 2023: The p.D117H variant (also known as c.349G>C), located in coding exon 4 of the MITF gene, results from a G to C substitution at nucleotide position 349. The aspartic acid at codon 117 is replaced by histidine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
