NM_000071.3(CBS):c.1484C>T (p.Thr495Met) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T495M variant (also known as c.1484C>T), located in coding exon 14 of the CBS gene, results from a C to T substitution at nucleotide position 1484. The threonine at codon 495 is replaced by methionine, an amino acid with similar properties. This variant has been detected in a psychiatric disorders cohort (Sriretnakumar V et al. Am. J. Med. Genet. B Neuropsychiatr. Genet., 2019 01;180:46-54). This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
