Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_004991.4(MECOM):c.3709A>G (p.Ser1237Gly), citing Ambry Variant Classification Scheme 2023: The p.S1237G variant (also known as c.3709A>G), located in coding exon 17 of the MECOM gene, results from an A to G substitution at nucleotide position 3709. The serine at codon 1237 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
