Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001267550.2(TTN):c.70243A>C (p.Met23415Leu), citing Ambry Variant Classification Scheme 2023: The p.M14350L variant (also known as c.43048A>C), located in coding exon 153 of the TTN gene, results from an A to C substitution at nucleotide position 43048. The methionine at codon 14350 is replaced by leucine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species, and leucine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
