Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001267550.2(TTN):c.19036G>A (p.Val6346Met), citing LabCorp Variant Classification Summary - May 2015. This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 19036, where G is replaced by A; at the protein level this means replaces valine at residue 6346 with methionine — a missense variant. Submitter rationale: Variant summary: TTN c.15304G>A (p.Val5102Met) results in a conservative amino acid change located in the I-band region of the encoded protein sequence. Three of four in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 2e-05 in 247020 control chromosomes. The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.15304G>A in individuals affected with Cardiomyopathy/Limb-Girdle Muscular Dystrophy/TTN-related disorders and no experimental evidence demonstrating its impact on protein function have been reported. Three clinical diagnostic laboratories have submitted clinical-significance assessments for this variant to ClinVar after 2014 without evidence for independent evaluation. Based on the evidence outlined above, the variant was classified as uncertain significance.
