This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 51037, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 17013 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change results in a premature translational stop signal in exon 270 of the TTN mRNA at codon 17013 (p.Glu17013*). While this is not anticipated to result in nonsense mediated decay, it is expected to create a truncated TTN protein. This variant is found in the A-band of this gene.
