This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 63332, where C is replaced by T; at the protein level this means replaces serine at residue 21111 with phenylalanine — a missense variant. Submitter rationale: Variant summary: TTN c.55628C>T (p.Ser18543Phe) results in a non-conservative amino acid change located in the A-band (https://www.cardiodb.org) of the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-06 in 247984 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. To our knowledge, no occurrence of c.55628C>T in individuals affected with Limb-Girdle Muscular Dystrophy, Type 2J and no experimental evidence demonstrating its impact on protein function have been reported.
